John Kennedy Toole's A Confederacy of Dunces (1980) is a standalone novel following Ignatius J. Reilly, a large, medieval-philosophy-obsessed thirty-year-old forced by his mother to seek employment in 1960s New Orleans. Published posthumously after Toole's suicide, it won the Pulitzer Prize in 1981 and is one of the funniest American novels ever written.
